PH1004 is a research-stage proposed biosimilar candidate referencing rituximab. The intended molecule is a recombinant chimeric murine/human IgG1 kappa monoclonal antibody directed against the CD20 antigen. Planned development activities include mammalian-cell culture process development and stepwise comparative analytical and functional studies. Biosimilarity has not been established.

Development Focus

The program is focused on comparative evaluation of molecular structure, glycan and charge profiles, purity, aggregates, stability, CD20 binding, Fcγ-receptor and C1q interactions, complement-dependent cytotoxicity, antibody-dependent cell-mediated cytotoxicity, and relevant B-cell lysis assays. The reference-product context includes CD20-positive B-cell malignancies and selected autoimmune or inflammatory disorders. These are not established indications for PH1004.

Proposed Mode of Action

PH1004 is designed to bind CD20 expressed on pre-B and mature B lymphocytes. Following target binding, the candidate is intended to recruit complement and immune effector cells, supporting complement-dependent and antibody-dependent mechanisms of CD20-positive B-cell lysis. These mechanisms remain subject to comparative functional confirmation.
