Scientific Profile

PH1003 is a research-stage proposed biosimilar candidate referencing bevacizumab. The intended molecule is a recombinant humanized IgG1 monoclonal antibody designed to bind vascular endothelial growth factor A (VEGF-A). Planned development activities include mammalian-cell culture process development and stepwise comparative analytical and functional studies. Biosimilarity has not been established.

Development Focus

Development is focused on comparative evaluation of molecular structure, glycosylation, charge and size variants, purity, aggregates, stability, VEGF-A binding, inhibition of VEGF-A interaction with VEGFR-1 and VEGFR-2, and endothelial-cell proliferation bioassays. Reference-product authorizations span several solid-tumor settings and vary by jurisdiction. None is an established indication for PH1003.

Proposed Mode of Action

PH1003 is designed to bind soluble VEGF-A and limit its interaction with VEGFR-1 and VEGFR-2 on endothelial cells. If confirmed through comparative functional studies, this activity would be expected to attenuate VEGF-A-mediated endothelial-cell proliferation and new-vessel formation in relevant experimental systems. These are proposed functional attributes and not established clinical effects of PH1003.
